Try running a G92 E0 command to reset the extruder before you try to extrude the 100mm for the calibration test. The G92 E0 command resets the extruder.
Cables are correctly connected, and the Rumba board does not have an on-board SD Memory Card slot. I will keep tooling around with this in my free time but since I use octopi it's not a massive deal....

1. When trying to fix an issue change ONE THING AT A TIME and then test that change to see if it helped.

Printed out a test cube, thing came out to within .03mm of spec. Having no issues with Marlin RC so far.
